Cross talks between the immune and lymphatic endothelial cells regulate inflammatory lymph node lymphangiogenesis: defining a new therapeutic approach

چکیده

Introduction The lymphatic system serves as the primary route for the metastasis of many cancers and the extent of lymphangiogenesis involvement is the most important indicator of tumor aggressiveness. Despite the apparent importance of the lymphatic vessels for tumor dissemination, it has remained unclear whether immune cells and lymphatic endothelial cells interactions may affect tumor progression and metastasis. Recent evidences indicate regulation of lymph node lymphangiogenesis by T lymphocytes. However, the nature of the T cells involved and the nature of the immune response in inflammatory lymph node lymphangiogenesis are still unveiled. We therefore characterized the nature of the inflammatory response in inflammatory lymph node lymphangiogenesis.

Lack of lymphatics and lymph node-mediated immunity in choroidal neovascularization.

PURPOSE Inflammation and immune cells regulate choroidal neovascularization (CNV) and could become therapeutic targets in age-related macular degeneration (AMD). Lymphangiogenesis is a key component of various inflammatory diseases. Whether lymphangiogenesis and lymph node-mediated immunity are involved in the pathogenesis of AMD is not understood. METHODS To investigate lymphangiogenesis in ...

CD4+ Cells Regulate Fibrosis and Lymphangiogenesis in Response to Lymphatic Fluid Stasis

INTRODUCTION Lymphedema is a chronic disorder that occurs commonly after lymph node removal for cancer treatment and is characterized by swelling, fibrosis, inflammation, and adipose deposition. Although previous histological studies have investigated inflammatory changes that occur in lymphedema, the precise cellular make up of the inflammatory infiltrate remains unknown. It is also unclear if...

The A2A adenosine receptor regulates the formation of new lymphatic vessels in inflammatory microenvironments

The formation of new lymphatic vessels, or lymphangiogenesis, is a natural process involved in tissue repair and in the resolution of inflammatory reactions. In some cancers, this process is exploited by the tumor to promote its growth and its metastatic dissemination. Our group recently identified a role for the CD73-adenosine pathway in tumor angiogenesis. As angiogenesis and lymphangiogenesi...

Recent developments in morphology of lymphatic vessels and lymph nodes.

This paper reviews the morphology of lymphatics and lymphangiogenesis in vivo, microenvironments that promote lymphangiogenesis, and the structure and function of lymph nodes. Lymphatic capillaries consist of a single layer of lymphatic endothelial cells (LECs) and have valves, while collecting lymphatics are endowed with smooth muscle cells (SMCs) and valves besides a single layer of LECs. In ...

The Lymphatic Endothelial mCLCA1 Antibody Induces Proliferation and Growth of Lymph Node Lymphatic Sinuses

Lymphocyte- and leukocyte-mediated lymph node (LN) lymphatic sinus growth (lymphangiogenesis) is involved in immune responses and in diseases including cancer and arthritis. We previously discovered a 10.1.1 Ab that recognizes the lymphatic endothelial cell (LEC) surface protein mCLCA1, which is an interacting partner for LFA1 and Mac-1 that mediates lymphocyte adhesion to LECs. Here, we show t...
